How do you clean the ravioli stamp before first use and after making pasta?

As new, it shouldn't need cleaning, other than maybe wiping the bottom surface with a moist towel. I wouldn't worry about it unless you see something like machine oil on it. In normal use, you shouldn't be seeing pasta getting stuck to it (or wedged inside). When done, just let it dry, and brush off any residual bits of pasta. If your dough has olive oil, a thin film might be there, but I haven't seen anything worth dealing with. I would avoid taking it apart because the threads are not designed for regular maintenance. If you have to, it can be taken apart. Just don't cross thread or over-tighten, and I don't know the recommended torques. To take apart, unscrew the handle. Unscrew the two nuts - ONE at a time. It's spring loaded, so just be aware. Don't use dishwasher, any chemicals that react strongly with copper, or abrasives.

Is the filling 50mm or edge to edge 50mm?

The cutter is 50 mm external diameter. The internal "plunger" (roughly, the maximum filling size) is 37 mm.
